The servants

Saber: Saber's master is Kiritsugu Emiya, with whom she is often at odds. She is also known as Artoria Pendragon, the King of Knights and King Arthur. She fights to obtain the Holy Grail so that she can rewrite the past to save her kingdom, Great Britain.
Archer: The archer's master is initially Tokiomi Tohsaka, but later Kirei Kotomine takes his place. He is also known as Gilgamesh and the King of Heroes. He sees the Holy Grail as a treasure that already belongs to him and only participates in the Holy Grail War for his own amusement and to prevent other servants, all those he despises, from stealing what he already considers his treasure.
Lancer: Lancer's master is Kayneth El-Melloi Archibald. He is also known as Diarmuid Ua Dubhne. He has no desire to obtain the Holy Grail; he only cares about being a knight by serving his master. Later in the series, when Kayneth El-Melloi Archibald is injured, he is given another reason to fight, which is to restore his health.
Rider: Rider's master is the young Waver Velvet. He is also known as Iskander, Alexander the Great, and King of Conquerors. The way he believes a king should behave is very different from Saber's, and they disagree on this subject. He fights in the Holy Grail War to begin reincarnation in the modern world.
Roulette: Caster's master is the serial killer Ryuunosuke Uryuu. He is also known as Bluebeard and Gilles de Rais. He is a sociopath who greatly enjoys murdering, especially children. He hopes that the woman he is obsessed with, Joan of Arc, will be resurrected by the Holy Grail. He mistakes Saber for Joan of Arc due to their similar appearance.
Berserker (Lancelot of the Lake/formerly the Black Knight): Berserker's master is Kariya Emiya. He is also known as Lancelot of the Lake or formerly the Black Knight. Having wronged Saber by having an affair with King Arthur's wife, he sought punishment from Saber to be forgiven for his misdeeds and be at peace.
Assassin: The master assassin is Kirei Kotomine. His personality has been split into several beings, and his wish is for them all to return to him so he can become whole again.

Main human characters

Emilie Kiritsugu: Kiritsugu Emiya specializes in eliminating hired mages. There is no weapon or strategy he doesn't employ. His profession has cost him everyone he loves. He wants the Holy Grail to grant his wish for world peace. His wife is Irisviel von Einzbern. He has a daughter with her named Illyasviel von Einzbern.
Irisviel von Einzbern: Irisviel von Einzbern's husband is Kiritsugu Emiya and her daughter is Illyasviel von Einzbern. She is not human, but rather a creation of the Einzbern family called a homunculus. She takes a humanoid form and possesses a consciousness that helps her defend herself, but she is actually the vessel for the Grail. She has led a very sheltered life in order to protect herself.
Kirei Kotomine: Kirei Kotomine's father is Reise Kotomine, the overseer of the Fourth Holy Grail War and a priest of the Church. He follows all of his father's orders and doesn't believe he has any wishes of his own. Archer eventually awakens his interest in understanding what he truly wants, and his wish becomes the Holy Grail, revealing it to him. He is preoccupied with Kiritsugu Emiya.
Tokyo Tohsaka: Tokiomi Tohsaka is the head of the Tohsaka family. He is a skilled combat mage who uses gem and fire magic. His wish is to use the Grail to reach the Akasha Root.
Aoi Tohsaka: She is the wife of Tokiomi Tohsaka and the mother of Rin Tohsaka and Sakura Matou. Aoi Tohsaka is married to Tokiomi Tohsaka. Her daughter is Rin Tohsaka. She also has another daughter named Sakura Matou whom she allowed to be adopted by the Matou family upon recommendation from her husband who wanted her to be trained as a mage by them.
Waver Velvet: Waver Velvet is a young student at the Clock Tower. He doesn't come from a strong or long line of mages. He's only third generation, so he's not very respected. His wish is for the Mage Association to recognize his genius. Despite this, he's often scared and has insecurities, but his Servant Rider helps build his confidence as the season progresses.
Maiya Hisau: Maiya Hisau is Kiritsugu Emiya's assistant. She always has a serious personality and follows all his orders. She doesn't do anything for herself, as we discover when she shares her mysterious past with Irisviel von Einzbern.
Kariya Matou: Kariya Matou cares deeply for Aoi Tohsaka and her daughters Sakura Matou and Rin Tohsaka. He makes a deal with her father Zouken Matou to have him undergo the brutal (possibly fatal) training required to participate in the 4th Holy Grail War in exchange for Sakura Matou returning to her family and allowing her to live a normal life.
Ryûnosuke Uryû: Ryûnosuke Uryû is a serial killer who is always looking for new ways to kill people. He has no wish for the Holy Grail but instead wishes to continue having fun with Caster as they improve their methods of murder.
Kayneth El-Melloi Archibald: Kayneth El-Melloi Archibald is a professor at the Clock Tower and a member of the Mages' Association. He participates in this activity to strengthen the respect he has among his peers.
Sola-Ui Nuada-Re Sophia-Ri: Sola-Ui Nuada-Re Sophia is married to Kayneth El-Melloi Archibald.
Risei Kotomine: Risei Kotomine's son is Kirei Kotomine. He is a church priest and oversees the 4th Holy Grail War, but also works with Tokiomi Tohasaka and Kirei Kotomine to ensure that the Holy Grail is claimed by Kirei Kotomine for the church.

Minor human characters

Rin Tohsaka: Rin Tohsaka's father is Tokiomi Tohsaka and her mother is Aoi Tohsaka. Her father teaches her gem magic.
Sakura Matou: Sakura Matou's father is Tokiomi Tohsaka and her mother is Aoi Tohsaka, but they gave her up to be adopted by the Matou family, showing that she would become a powerful mage under Zouken Matou's training.
Illyasviel von Einzbern: Illyasviel von Einzbern's father is Kiritsugu Emiya and his mother is Irisviel von Einzbern
Jubstachit of Einzbern: Head of the Einzbern family and grandfather of Illyasviel von Einzbern.
Matou Zouken: Head of the Matou family. He is a cruel and merciless mage who mercilessly trains Sakura Matou. His son is Kariya Matou.
Glen and Martha Mackenzie: Waver Velvet cast a spell on this couple to make them think they were his grandparents. They let him and Rider stay at their house.
Norikata Emiya: Kiritsugu Emiya's father who was researching the Dead Apostles.
Shirley: Kiritsugu Emiya's childhood friend.
Natalia Kaminiski: She raised Kiritsugu Emiya and taught him her ways as a freelance hitman.
Shirō Emiya: Kiritsugu Emiya's adopted son.
